One of the most importorpant problems in a direct analysis by the atomic absorption spectrometry with solid sampling technique is an establishment of the preparation of calibration curves which are reliable on the analytical accuracy.We have tried to use an internal standard method which is useful for the compensation of variation of base line. However, this method has not brought out a particular improvement in the analytical accuracy and linearity of calibration curves, but this method gave some interesting results for the direct determination of lead and selenium by atomic absorption spectrometry with solid sampling technique after preconcentration of them by using APDC-Ni coprecipitation.We have used standard solutions to prepare the calibration curves for the direct determination of lead, zinc etc. in biological samples, and analytical results for these volatile elements are in satisfactory agreement with the certified values or other method.We have also tried to prepare the calibration curves by using a chelate coprecipitation with magnesium 8-quinate as synthetic standard materials for solid sampling technique with atomic absorption spectrometry, and it was found that the synthetic standard materials were also useful.In this work, the standard addition method for solid sampling technique has been established for the direct determination of nickel, cobalt, copper and manganes etc. at the ppm levels in powdered biological samples, and it has been found that this new preparation method of calibration graphs was the most reliabe on the analytical accuracy.
