Pfefer et al等の方法(Investigative Ophthalmlogy and Visual Science,27:1031-1040,1986)を改変し,0.1mMカルシウム下で牛胎児血清を1%にした無血清培養に近い方法で行い,in vitroでin vivoに近い培養網膜色素上皮細胞を再構成した。アクチンについてはロ-ダミン標識のphalloidinを用いて調べた.アクチンはニワトリの網膜色素上皮細胞で報告されているのと同様,細胞の側壁に分布するcircumferential microfilament bundlesと微絨毛に存在するparacrystaline bundlesとして分布していた.中間型フィラメントとしてはビメンチンをウエスタンブロッティング方法で確認し,細胞内にメッシュ状に分布することをつきとめた.ケラチンは同定されなかった.チューブリンは細胞質内にわずかに存在していた.網膜色素上皮細胞は人においてもケラチンを有さない特異な上皮細胞であることがわかった.
