STD系ラット雄性を麻酔後、開胸、左冠状動脈の起始部の下方で結紮、すみやかに閉胸した。結紮後、それぞれ0分、5分、10分、15分、20分、30分、45分、60分後に麻酔下で大動脈切断により出血死させた(各群とも3匹使用)。心臓を摘出、ホルマリン固定後、3μmに薄切し、シラン処理スライドガラスに貼付し、抗ユビキチン抗体およびHSP72を染色することが判明している抗熱ショック蛋白抗体を用いて免疫染色を行った。結紮後20分以上の標本については、抗ミオグロビン抗体で心筋虚血を確認した。正立型落射蛍光顕微鏡にビデオカメラを取り付け、ステージ部分にオートスキャニング装置を組み込み、これらが同調するようにプログラムされた自動顕微測光装置を用いて、ピクセル直径40μmの円内の測光(1cm^2あたり20×20に分解)を行った。この装置により虚血部位と非虚血部位の光強度を測定してそれぞれの染色強度の平均値を求めてその比を出し(虚血指数)、0分群と各時間群との有意差の検定をStudent's t-testにより行った。その結果、抗ユビキチン抗体染色では15分以降で、有意差があり、抗HSP72抗体染色では60分以内では有意差は認められなかった。ユビキチンは虚血早期から出現することが判明し、急性期の心筋虚血障害の病理学的診断の一助となることが明らかとなった。HSP72については今回の実験では有意差はなかったが、経時的な虚血指数の変化は明らかであり、今後検討する必要があるものと思われた。

STD is male, thoracic, left coronary artery ligation below the origin, chest closure. After ligation, the blood was cut off at 0, 5, 10, 15, 20, 30, 45, 60 minutes after ligation (3 for each group). After the heart was extracted, the protein was fixed, and the protein was thinly cut at 3μm. The protein was stained with anti-protein antibody and HSP72. More than 20 points after ligation, anti-ligated antibodies and heart muscle deficiency were confirmed. Vertical epi-optic micro-mirrors are used to measure light within a range of 40μm in diameter (1cm^2) and 20×20 in resolution). The light intensity of the device is measured at the empty blood part and the non-empty blood part, and the average value of the staining intensity of the device is calculated. The ratio is calculated (empty blood index). The time group is divided into 0 groups. The intentional difference is determined by Student's t-test. Results: Anti-HSP72 antibody staining was less than 15 points, intentional difference was less than 60 points, intentional difference was less than 60 points The pathological diagnosis of heart muscle deficiency and blood disorder in acute stage is helpful to the early diagnosis of heart muscle deficiency and blood disorder. HSP72 is a very important indicator of the health of the elderly.
