血球細胞であるリンパ球の皮膚への浸潤は皮膚におけるすべての炎症性疾患、腫瘍性疾患を研究するうえで欠くべからざるものであるが、そのメカニズムは解明されていない。なかでも皮膚T細胞性リンパ腫における表皮向性(epidermotropism)は有名であるが、なぜ皮膚リンパ腫細胞がこの性質を持つかはいまだ不明である。今回我々は、さまざまな浸潤形式を持つ皮膚リンパ腫において、まず免疫組織学的にin vivoでリンパ腫細胞がどのような接着分子(インテグリン、イムノグロブリンスパーファミリー、カドヘリン等)を発現しているかを検討し、さらにフローサイトメトリーを用いてこれらを確認した。その結果、これまで知られていた表皮細胞上のICAM-1とリンパ球上のLFA-1による結合のみではリンパ球の表皮内浸潤のすべてを説明できないことを判明した(菊地新他、第19回日本研究皮膚科学会にて発表)。つまり表皮内浸潤をともなう皮膚リンパ腫においてかならずしもICAM-1とLFA-1の両者が発現されておらず、この過程におけるその他の分子の関与が示唆された。またその際、超微細局在を確かめるため、post-embedding immunogold法(電子顕微鏡下)を用いて検討したところ、これらの接着分子が細胞膜表面に存在することが証明された。その後、皮膚リンパ腫細胞から抽出したm-RNAと設定した特殊なプライマーを用いRT-PCR法でこれら接着分子の検索をおこなった。その結果、100-150bpのPCR産物を得、これらを精製した後クローニングを行い、現在シークエンス施行中である。

The expression of ICAM-1 and LFA-1 in the epidermis is related to the expression of other molecules in the epidermis. The presence of adhesion molecules on the surface of the cell membrane was demonstrated by the post-embedding immunogold method (under electron microscope). After treatment, the skin cells were extracted from the m-RNA set and the molecular markers were detected by RT-PCR. Results: PCR products of 100-150bp were obtained, purified and processed.
