本研究では、アルカリ外傷角膜におけるIL-8の発現について検討した。まず、家兎角膜アルカリ外傷におけるInterleukin8(IL-8)の発現についてRT-PCR法を用いて検討した。正常およびアルカリ外傷後7日の角膜を摘出、AGPC法を用いてtRNAを抽出しreversetranscriptionによってcDNAを作成した。プライマーはrabbitIL-8のcDNA塩基配列をもとに特異的プライマーを独自に作成した。アルカリ外傷は家兎角膜に1N NaOHを浸透させた直径10mmのろ紙を30秒間接触させて作成した。その結果、IL-8cDNAのサイズに一致するバンドをアルカリ外傷角膜にのみ認め、アルカリ外傷角膜においてIL-8 mRNAが発現していることが確認された。次にELISA法を用いてアルカリ外傷角膜におけるIL-8発現についてタンパクレベルでの定量を行った。1995年7月にラットのIL-8 ELISA systemが市販されたため、ラット角膜アルカリ外傷モデルを用いて実験を行った。ラット角膜28眼に1N NaOHを浸透させた直径5mmのろ紙を60秒間接触させてアルカリ外傷を作成した。無処置の正常角膜4眼を対照とした。正常角膜および外傷後1、3、5、7、14、21、28日の角膜を摘出、凍結・粉砕したのちPBSに浮遊させ遠心し、その上清についてIL-8の定量を行った。その結果、正常角膜において53.8±9.7(mean±s.e.)pg/corneaのIL-8発現を認め、外傷後1日には295.6±46.3pg/coernea、外傷後5日に最大値455.8±124.2pg/coerneaに達した。しかし、外傷後14日のIL-8発現は18.7±2.1pg/coerneaと正常角膜の約3分の1のレベルであり、外傷後28日においても外傷後14日と同じレベルであった。以上より、(1)正常角膜において約54pg/corneaのIL-8が発現する。(2)アルカリ外傷角膜では外傷後早期にIL-8が発現誘導され、外傷後5日に正常角膜の約8.5倍になる。(3)アルカリ外傷後14〜28日の瘢痕化角膜ではIL-8発現が正常の約1/3であることが明らかとなった。現在、角膜におけるIL-8の局在を免疫組織化学的手法を用いて検討中である。

The normal cornea was placed in 4 eyes. On the 1st, 3rd, 5th, 7th, 14th, 21st and 28th day after the normal cornea was removed, the cornea was removed, the powder was removed, the PBS was floating, the heart was floating, and the supernatant was quantified by IL-8. The results showed that the normal cornea was abnormal (pg/ ±s. E.). Il-8 showed abnormal symptoms, 295.6 ±46.3pg/coernea on the 1st day, and 455.8 ±124.2pg/coernea on the 5th day after surgery. On the 14th day after surgery, the IL-8 showed that the normal cornea was 18.7 ±2.1pg/coernea, and the normal cornea was normal. The results showed that: (1) the normal cornea showed abnormal expression of IL-8 in 54pg/ cornea. (2) the normal cornea was 8. 5 times higher than that of the normal cornea in the early stage after IL-8 surgery and 5 days after surgery. (3) from 14 to 28 days after surgery, cicatricial IL-8 was found to be normal. At present, the Corneal Corneal tissue IL-8 Bureau is using immunocytochemical techniques.
