Cytochrome P4502E1 (CYP2E) is a member of the microsomal ethanol oxidizing enzymes, and it is well-known that CYP2E is induced when ethanol is the consumed chronically or largely. There have been, however, few reports to evaluate the extent of induced CYP2E1 level. The aim of this study was to establish the method for evaluating CYP2E1 mRNA level and to examine the correlation between the expression level of CYP2E1 mRNA and the individual difference of susceptibility to ethanol by this method.It was possible to measure the level of CYP2E1 mRNA in human blood lymphocyte by Reverse Transcriptase-PCR (quantitative RT-PCR method). There were no differences between the healthy individuals without drinking and with habitual drinking in CYP2E1 mRNA levels using quantitative RT-PCR method. Moreover, there were also no differences of the expressed levels of CYP2E1 mRNA in each of the three genotypes of ALDH2 when various concentrations of ethanol were added in the blood and incubated at 37℃ for a definite time in vitro.It was suggested that the drinking frequency or the strength of ALDH2 activity has no effect on the expressed levels of CYP2E1 mRNA.And we studied the relationship between allelic frequencies of alcohol metabolizing enzymes (ALDH2, ADH2, ADH3, and CYP2E1), and yearly per capita consumption of alcohol regionally. It was observed that only ALDH2*1 gene frequency was the lowest in the center of Japanese island (Kinki, Tokai, and Hokuriku region). But there were no significant correlations between the regional yearly per capita consumption of alcohol and ALDH2*1 gene frequency.
