ヒト結腸におけるc-kitとMITFアイソフォームの発現をヒト結腸から抽出したトータルRNAを用いてRT-PCRを行い検討した。結腸においてはMITFアイソフォームであるMITF-AとMITF-H mRNAの発現は認められたが色素細胞特異的に発現しているMITF-M mRNAは検出されなかった。またc-kit mRNAの発現は認められた。ヒトc-kit遺伝子のプロモーターをルシフェラーゼ遺伝子の上流に組み込んだベクターとMITFアイソフォームの発現ベクターを作製し、c-kit遺伝子プロモーターに対するMITF アイソフォームの転写活性化能をヒト結腸癌由来のDLD-1、ヒト子宮頚癌由来のHeLa、黒色腫由来のHMV-IIを用いてtransient cotransfection assayを行い検討した。しかし、この系ではっきりとしたc-kit promoterに対する転写活性化能を検出し得なかった。続いて肥満細胞由来のHMC-Iを用いてtransient cotransfection assayを行い検討した結果、MITF-Aは少なくとも同プロモーターに対して転写活性化能を有する傾向が見られた。これまでの実験から少なくとも結腸において発現、機能しているアイソフォームはMITF-AもしくはMITF-Hであるという結果が得られた。また肥満細胞由来のHMC-1のみでしか確認できていないが、c-kit promoterに対してMITF-Aは転写活性化能を有していることが確認された。しかし、MITF-HにはAのような転写活性化能は確認できなかった。

C-kit and MITF were used to detect RNA from the colon. MITF-A and MITF-H mRNA expression in the colon was detected in pigment-specific cells. The expression of c-kit mRNA was detected. C-kit gene expression profile analysis and mitf gene expression profile analysis were performed to investigate the origin of colon cancer DLD-1, uterine cancer HeLa, and melanoma HMV-II using the transient cotransfection assay. This is a c-kit promoter for the activation of a cell phone. The HMC-I assay was used to investigate the origin of mitotic cells. The MITF-A assay was used to investigate the activity of mitotic cells. This is the first time that a person has ever been involved in a crime. HMC-1 was identified as the origin of mitotic cells, and the c-kit promoter was identified as the activity of mitf-A. MITF-H can be used to activate A cell.
