项目摘要

The purpose of the present investigation was to determine, by means of the ELISA, whether syngeneic mouse serum (SS) becomes antigenically altered when conbined with various medicaments. Syngeneic sera modified with six agents were used as the immunogens to induce antibody production in BALB/c mice. Six agents used in this investigation were tincture of iodine, formocresol, formalin guaiacol, neutral formalin, meta-cresol and guaiacol. Immunogenicity could be conferred upon tolerated syngeneic serum by modification with tincture of iodine (J) or formocresol (FC) or formalin guaiacol (FG). However, it is suggested that the synergistic action of formalin and cresol or guaiacol is an indispensable condition for the acquirement of immunogenicity of FC-modified or FG-modified syngeneic serurn (FC-SS, FG-SS). Moreover, the immunological tolerance to native syngeneic serum is not terminated following immune-injection of J-SS or FC-SS or FG-SS.Next, the levels of each class and subclass of immunoglobulins (IgG1, IgG2a, IgG2b, IgG3, IgE, and IgA) in serum of BALB/c immuned by J-SS or FC-SS or FG-SS were measured. The increased production of IgG1, IgG2b, IgG2a and IgE in serum obtained from mice sensitized with J-SS were confirmed. The levels of IgG1 in antiserum to J-SS increased at about 27-th times compared with control, and incresed in order of IgG2b, IgE, IgG2a. The levels of IgG1 in antiserum to FC-SS increased at about 18-th times compared with control, and incresed in order of IgG2a, IgG2b The levels of IgG1 in antiserum to FG-SS increased at about 13-th times compared with control, and incresed in order of IgG2a, IgG2b.
