今回の科学研究費で、漢方薬による眼炎症性疾患に対する基礎的研究を行った。実験動物には有色家兎を用い、インターロイキン1の硝子体内注射で惹起した眼内炎症に対する漢方薬(黄連解毒湯・オウゴン)の抑制効果を調べた。漢方薬含有飼料を家兎に投与した。眼炎症の指標として、前房内フレア値を経時的に測定し、コントロール群と比較・検討した。結果として、黄連解毒湯及びオウゴン投与群はコントロールと比較し、有意に前房内フレア上昇を抑制した。黄連解毒湯及びオウゴンはインターロイキン1で惹起した眼内炎を抑制することを明らかとした(American Journal of Chinese Medicine, in press)。オウゴン以外の黄連解毒湯に含有される生薬であるオウバク・オウレン・サンシシとサンシシの含有成分であるcrocetinを用いて有色家兎で、リポポリサッカライド(LPS)の全身投与及びプロスタグランジンE2の角膜浸透法で惹起した眼内炎症抑制効果の研究を行った。結果として、サンシシ及びサンシシの含有成分であるcrocetinがLPSで惹起した眼内炎を有意に抑制することを明らかとした(投稿中)。オウゴンに含有される化学成分である、baicalein, baicalin, wogoninの、ヒト網膜色素上皮の培養細胞をインターロイキン1βで刺激して産生される、インターロイキン6、8のmRNA発現の抑制効果の研究を行った。wogoninが有意にインターロイキン6、8 mRNA発現を抑制することを明らかとした(投稿中)。この基礎的研究によって眼炎症性疾患に対する、漢方薬を用いた新しい治療法を発見した。
