本研究は変異単純ヘルペスウイルス(HSV-1716)の経気管支投与による抗腫瘍効果および安全性を多発性肺腫瘍マウスモデルを用いて検討するものである。1.腫瘍内における変異HSVの存在の確認担癌BALB/Cマウスに4X10^6pfuのHSV-1716を経気管支的に投与後、両側肺を摘出した。両側肺内に悪性腫瘍の存在を確認後、抗HSV抗体による免疫組織染色を行った。腫瘍内に比較的選択的に染色を認め、HSV-1716の腫瘍内における選択的増殖を確認した。2.経気管支的投与後における変異HSVの体内分布の検討EJ-62多発性肺腫瘍マウスモデルに対し経気管支的にHSV-1716を投与し、標的臓器である肺以外の臓器(脳、脊髄、肝、腎、心臓、腸管、皮膚、眼球)を摘出した。HE染色による病理組織標本の観察では経気管支的治療に伴う明かな形態的学的変化を認めなかった。抗HSV抗体による免疫染色でも非標的臓器における明らかな陽性像は認めなかった。HSV thymidine kinaseを目標としたPCRも実施したが皮膚で少量の感染を検出した以外はいずれも陰性であった。3.血清酵素の測定治療群と無治療群の肝機能障害を比較するために血清AST、ALT、LDHの比較をおこなった。今回のHSV-1716投与量においては両群間に有意差を認めなかった。

This study investigated the effects of HSV-1716 on anti-tumor and safety in patients with multiple lung cancer. 1. The presence of HSV-1716 in BALB/C cells was confirmed after administration of HSV-1716 to the peripheral blood vessels. After confirmation of the presence of HSV lesions in the lungs, anti-HSV antibodies were used for immunohistochemical staining.
