マウス卵巣摘出モデルを用いたTyro3,Gas6の検討:8週令雌ddyマウスを偽手術群と卵巣摘出群(n=6)にわけあわせてエストロゲンの効果も検討する為、手術直後に卵巣摘出群にエストロゲン(3-week-release;0.01mg/p)とプラセボを、偽手術群にプラセボの錠剤を皮下に埋入し、3週後に子宮及び脛骨を摘出した。脛骨は、骨髄と残りの骨とにわけ、それぞれ、mRNAレベルにおけるTyro3とGas6の発現をNorthern blotで検討した。また、破骨細胞の分化や活性、延命に深くかかわっていることが明らかになっている他の受容体型チロシンキナーゼとそのリガンド(FGFR1 & FdF-2,c-Fms & M-CSF)についても骨髄細胞と骨における発現をNorthern blotで検討した。骨髄細胞におけるGas6のm-RNAレベルでの発現は、卵巣摘出群で偽手術群よりも増加していたが、エストロゲン投与によってその増加は抑制された。一方Tyro3発現は、脛骨と骨髄細胞ともに見られなかった。しかし、同じRNAを用いてRT-PCRを行った結果、脛骨では卵巣摘出群、偽手術群のいずれにおいてもTyro3の発現が見られた。このことから、今回回収されたRNAが微量なために、Northern blotではTyro3が検出できなかったと考えられる。いずれにしても今回の検討から、卵巣摘出による骨粗鬆症の病態にGas6の発現が関与している可能性が示唆された。
