OPG(osteoprotegerin)添加溶液の歯牙用保存液としての効果を判定するために、生後4週齢ラットの上顎第一臼歯を用いて再植実験を行った。全身麻酔下のもと、抜去した右側第一臼歯を0.1,10,1000ng/mlに調整したOPG溶液中に約10分間保存した後、直ちに再植を行った。対照群には保存液として生理食塩水を用いた。再植処置後14日目に治癒の良否が組織学的に判定可能であることが確認された。対称群(生理食塩水)と比較して、OPG0.1ng/ml群では、治癒の良否に明らかな差は認められなかった。しかしながら、OPG10ng/ml群および1000ng/ml群では明らかに治癒良好例の割合が増加した。以上の結果から、OPG(OCIF)添加溶液は、外傷などで脱落した歯牙を保存するのに、有用であることが示唆された。5週齢のOPG遺伝子欠損マウスを用いた再植ならび脱臼を行った。全身麻酔下のもと、上顎第一臼歯を抜去し直ちに再植を行った(再植群)。また、脱臼操作も同歯牙に対して行った(脱臼群)。処置後1,3,7,14,28日目に組織学的観察を行った。対称群として野生型マウスで同様の処置を行った。処置後1,3日目には、OPG遺伝子欠損マウスの再植群ならび脱臼群共に、歯根を吸収する破歯細胞が出現し、対処群に比較して著しく増加しているのが観察された。14日目には歯根の内部からも吸収されている像も観察され、歯牙全体として大きく吸収を受けていた。28日目には歯牙がほぼ吸収されており、骨組織に置換されていた。以上の結果から、OPGの欠損によって、外傷などによって歯根膜に傷害が生じた場合、破歯細胞が出現し、同細胞によって歯根吸収が行われ、骨組織に置換されることが組織学的に観察された。損傷した歯根膜細胞はRANKLを発現して破歯細胞形成を促進する可能性、また、OPGは傷害を受けてRANKLを発現するようになった歯根膜細胞を抑制し、破歯細胞形成を抑制する可能性がそれぞれ推察された。

OPG(osteoprotegerin) solution was added to the dental preservation solution to determine the effect of replantation of the maxillary first tooth at 4 weeks after birth. Dislocation is the same as tooth movement (dislocation group). Histological observations were made on the 1st, 3rd, 7th, 14th and 28th day after treatment. The wild type is the same as the wild type 1, 3 days after treatment, OPG gene damage, replantation, dislocation, tooth root absorption, tooth damage, tooth damage. On the 14th, the tooth root and the tooth root were absorbed by the image, and the tooth was absorbed by the whole tooth. On the 28th, the teeth were replaced by bone tissue. The above results show that the defect of OPG, the injury of tooth membrane, the occurrence of tooth cell, the absorption of tooth tissue and the replacement of bone tissue are observed histologically. The possibility of RANKL expression and cell formation promotion in damaged dental root membrane cells, and the possibility of RANKL expression and OPG injury inhibition in damaged dental root membrane cells and cell formation were investigated.
