神経芽細胞腫は小児特有のがんで、近年、集学的治療が発達してきたにも関わらず、その予後は不良である。その発症は、胎生期に生じた分化関連遺伝子の発現変異あるいは翻訳された蛋白質の機能異常が蓄積した結果とも考えられる。神経芽細胞腫の発生は、交感神経節細胞と副腎髄質細胞に由来する。この両者の細胞は神経堤細胞から分化する。近年、これら神経細胞の発生に関与するいろいろな神経発生分化因子が、同定されてきた。神経発生分化因子として、BMP (bone morphogen protein)、MASH1、Phoxなどの転写因子があげられる。さらにこれらの下流には、神経栄養因子の受容体であるGDNF受容体、NGF受容体が存在する。特に下流の遺伝子群は神経芽細胞腫の予後を決定する上で非常に重要な因子である。このような神経発生分化の側面から、神経芽細胞腫の分化を促す研究も必要と思われる。レチノイン酸耐性株にレチノイド化合物Am80を添加し、下記の項目について検討する。1.培養細胞株の形態学的変化(分化の有無について)2.神経発生分化に関係する因子の発現(Nothern blot法)3.癌遺伝子、癌抑制遺伝子の発現(Northern blot法およびWestern blot法)その結果、形態学的に分化傾向がみられた。また神経発生分化因子の発現上昇も確認できた。レチノイド化合物Am80は、神経芽細胞腫の分化誘導剤として有望と考えられる。
