The Role of CEACAM1 (CEA-related cell adhesion molecule 1) in vascular morphogenesis and stabilization

Angiogenesis is regulated by a net balance of angiogenic activators and inhibitors. Cell adhesion molecules are also involved in the different stages of angiogenesis, in particular of vascular morphogenesis. We recently charakterized soluble CEACAM1 (CEA-related cell adhesion molecule 1) as a pro-angiogenic factor and morphogenic effector of VEGF. Increased CEACAM1 expression was found in endothelial cells of new blood vessels but the influence of membrane-bound CEACAM1 on the expression of known angiogenic factors and the morphogenesis of capillaries is still unknown. The aim of this study is to explore the role of membran-bound CEACAM1 in angiogenesis using the following experimental models: 1. Adenoviral overexpression of CEACAM1 in vascular endothelial and smooth muscle cells which will be subsequently used a) for gene array analyses regarding the expression of known potent angiogenic factors, b) migration and in vitro wound assays and c) endothelial tube formation assay. 2. Local microinjection of CEACAM1-adenovirus in chicken chorioallantoic membrane to study the effects of gene delivered CEACAM1 in angiogenesis and vascular leakage. These experiments will provide us with new insights concerning the effects of membrane-bound CEACAM1 on the angiogenesis.
