ラット切歯歯髄を歯軸に対して垂直方向に輪切りにし、象牙芽細胞層を含んだ歯髄組織切片を作製した。トリプシンとコラゲナーゼによる酵素処理を行ったあと、10%FCS添加培地にて30%窒素環境で培養を行った。培養日数によっては途中で細胞が脱分化してしまう恐れがあるが、explant tissue sampling、培養液の組成を含む至適培養条件の設定、培養日数について予備実験を行った。その上でガラスボトム35mm culture dish上で固定後TJタンパクに対する特異抗体であるclaudin-1、occludin、ZO-1を用い、それぞれ分化段階の違いによるTJタンパクの発現を観察した。経時的観察により培養象牙芽細胞は3日間で細胞形態を変化させていった。1日目では細胞長径と細胞突起が伸長し、核の偏在化が観察された。2日目では隣同士の細胞が接着しながら配列し、細胞突起は同一の方向を向くようになった。3日目には細胞の極性は失われ、上皮様細胞の像を呈するようになった。そこで培養1日目、2日目、3日目の象牙芽細胞においてTJ関連タンパクの発現を観察した。その結果、ZO-1は培養初期から細胞接着面に発現し、極性を失い上皮様を呈するようになっても強い発現を維持した。一方occludinは培養3日間を通して核のみに局在し細胞間での発現は観察されなかった。claudin-1は培養1日目には核内全体に発現を認めたが、2日目には核膜周域に散在するようになった。また細胞接着面には発現を認めなかった。

A side occludin is cultured for 3 days, and the nucleus is detected in the intercellular space. claudin-1 was cultured for 1 day and found in the whole nucleus, and for 2 days, it was found in the periphery of the nucleus. The cell surface is exposed to light.
