我们制作了HD-Ad.AAV。包含BTK外显子6~外显子19、TIMM8A和GFP-Hyg的BTK载体。用该载体处理雄性前b ALL细胞系Nalm6后，获得抗hyg克隆。Southern印迹法证实克隆中存在BTK基因重组。此外，将载体基因组整合到来自男性脐带血的cd34阳性细胞中。
英文摘要
We have made a HD-Ad.AAV.BTK vector containing BTK exon6～exon19, TIMM8A and GFP-Hyg. After treatment of a male pre-B ALL cell line, Nalm6, with the vector, Hyg-resistant clones were obtained. Recombination of BTK gene was demonstrated in the clones by Southern blotting. In addition, vector genome was integrated to CD34-positive cells from male cord blood.
