膵胆管合流異常における胆道上皮の発癌機序は解明されていない。我々はRNAレベルの網羅的な遺伝子発現を小児の合流異常の胆嚢上皮で検討し病態と関連する数種の遺伝子を同定することができた。なかでもUCA1(urotheial cancer associated 1)は、蛋白をコードしないnon-cording RNAにも関わらず、胎児と膀胱癌組織に発現して機能するoncofecal geneであり、合流異常の癌化に関与している可能性を考える。今回の研究では、このUCA1を成人期の正常胆嚢、胆道癌組織で合流異常の有無別にreal-time RT-PCRにて異常発現がないか解析し、合流異常による胆道癌発癌過程でのUCA1の役割を明らかにすることを目標としている。平成25年度はUCA1が胆道癌の発生に関与しているのかを調べるため、現在使用可能な状況となっている成人の非合流異常胆管癌の標本を用いた研究を行った。成人の非合流異常胆管癌の標本6検体を用い、各検体の癌部と非癌部からtotal RNAを抽出しcDNAを作成した。UCA1のmRNA発現量をreal-time RT-PCR法にて測定した。相対定量の結果からUCA1の発現量は、標本1の非癌部を1とした場合、標本2～6の非癌部の発現量はそれぞれ2.64、0.71、1.64、0.58、2.71であった。標本1～6の癌部の発現量はそれぞれ2.12、2.99、5.04、1.70、1.07、1.32であった。非癌部と癌部の発現量を統計学的に比較したところ有意差はでなかった。
