项目摘要

昨年度の研究の進展としては、1)去年採取できなかったレブンアツモリソウの種子サンプルを採取し、調査地の個体マッピングを完了した、2)レブンアツモリソウ種子サンプルからのゲノムDNAPCR法を確立した、以上2点が挙げられる。1)に関しては、昨年の天候不順によるレブンアツモリソウの不稔によるサンプル不足を補うことができた。また調査地の全個体の位置をマッピングし、二世代分析に用いる準備は整った。2)に関しては、Thomson & Henry(1995)によるDNA分離法とShimura & Koda(2004)による種子休眠打破法を組み合わせて用いることによって、ラン科種子のゲノムDNAからPCRを行うことに成功した。ラン科の種子は非常に微細なため、これまでリボソームDNAでの成功例はあったものの、ゲノム全体を対象としたPCRに成功したのはおそらく世界で初めての例である。これにより、ラン科における遺伝学的な研究に弾みがつくことを期待している。現在、論文をまとめている最中で、早ければ本年5月中には投稿できる予定である。残念ながら、昨年度から取り組んでいたマイクロサテライトマーカーを開発することはできなかった。さまざまな試行を繰り返し、試作したプライマーは数百に及ぶが、マーカーとして使用できたものはなかった。マイクロサテライトマーカーを作成することができれば、その後の二世代分析に用いるサンプルおよびデーターが揃っていただけに、マーカー作成の失敗が悔まれる。
