项目摘要

本研究は鉄過剰モデルマウスに対して,瀉血療法や鉄キレート療法など除鉄治療を行うことによる生体内での鉄代謝調節因子であるヘプシジンの変動や,各種鉄代謝関連遺伝子群の連動性,除鉄メカニズムを明らかにすることを目的に検討を行っている.iron dextranを腹腔内投与した鉄過剰モデルマウスに対して,1)未治療(鉄過剰群),2)鉄キレート剤のdesferrioxamine(DFO)投与群(DFO群),3)新規鉄キレート剤であるdeferasirox(DFX)少量投与群,4)DFX大量投与群,さらに5)未処置マウス(正常群)に分けて血液データの変化,肝臓・十二指腸・脾臓など病理組織の変化,各臓器における鉄代謝関連遺伝子群のmRNAレベルでの変化,蛋白レベルでの変化について解析を行っている.現時点までの検討では,血球数,肝機能に関しては各群間で変化はなく,血清鉄・フェリチン値は鉄過剰群と比べDFX群で低下が認められた.また血清中ヘプシジンのタンパク定量も鉄キレート療法群において減少傾向が見られた.定量的RT-PCR法によるmRNAレベルでの検討では,肝臓において鉄過剰群に比べ鉄キレート群においてTfR1をはじめ,TfR2,HFEにも除鉄治療により変化が見られ,また肝臓・脾臓においても鉄キレート群でFPN1mRNAの低下が認められ,鉄動態への影響が見られた.現在蛋白レベルでの鉄代謝関連遺伝子群の変化の解析を進行中であり,次年度前半での論文投稿を予定している.

Quantitative RT-PCR analysis of FPN1 mRNA expression in the liver, liver, spleen, and liver was performed in comparison with TfR1, TfR2,HFE and iron treatment. The analysis of the transformation of iron metabolism-related gene groups in protein metabolism is in progress, and the paper submission in the first half of the next year is scheduled.
