项目摘要

後肢懸垂モデルでのsoleus muscle萎縮時には低分子量熱ショックタンパク質(small heat shock protein, sHSP)の一つであるalpha B-crystallinが減少し、正常筋抽出物中においてαB-crystallinとtubulin/微小管が相互作用し、C2C12培養細胞系を用いてalpha B-crystallinの発現減少により筋管融合が起こらず微小管が正常に形成されないことが明らかとなっている(Takashi S et al., FASEB J, 19,1199-1201,2005.)。微小管は一般的細胞のみならず骨格筋細胞内において物質輸送などの重要な役割を果たすと共に細胞の形を保つための役割を担うと考えられている。骨格筋は力発揮のために構造が保たれることが必須であるが、ミクロなレベルで構造を保つために必須の役割を担う微小管に着目した骨格筋の研究はほとんどない。本年度の成果として、sHSPの一つであるalpha B-crystallinと微小管の構成成分であるtubulin,そして骨格筋の収縮タンパク質の一つであるmyosin heavy chainの骨格筋での相関性に関する論文を投稿論文として発表した。また、本科研費でalpha B-crystallinの抗体を作成し終えることが出来たため、今後もalpha B-crystallinの骨格筋細胞内での役割についての研究を続けていく予定である。
