Bei der neuronalen Exocytose bildet das vesikuläre SNARE-Protein Synaptobrevin-2 Komplexe mit den membranständigen SNAREs Syntaxin 1A und SNAP-25, wodurch die Fusionsreaktion eingeleitet wird. SNARE Proteine bilden nicht nur Komplexe untereinander, sondern sie bilden auch Homo-Oligomere, und außerdem sind zahlreiche weitere Bindungspartner bekannt. Um die räumliche Anordnung von SNAREs und SNARE-bindenden Proteinen zu untersuchen, haben wir ein Verfahren zur Herstellung von Plasmamembran-Rasen aus PC12-Zellen entwickelt. Diese Membran-Rasen behalten ihre Fähigkeit zur regulierten Exocytose, und sie sind für extern zugesetzte Reagenzien zugänglich. In diesen Membranen bildet Syntaxin Cholesterin-abhängige Mikrodomänen, die sich von Rafts unterscheiden und die Andock- und Fusionsstellen für sekretorische Granula definieren. Im hier vorgelegten Forschungsprojekt verfolgen wir zwei Ziele. Erstens soll die erhöhte Auflösung der 4Pi-Mikroskopie ausgenutzt werden, um den Abstand von sekretorischen Vesikeln zu den Syntaxin-Mikrodomänen mit einer Genauigkeit von 10-20 nm zu bestimmen und um diesen Abstand mit funktionellen Parametern zu korrelieren. Zweitens soll herausgefunden werden, ob Liposomen, die SNAREs sowie akzessorische Proteine enthalten, an solche Mikrodomänen binden und mit ihnen fusionieren können. Summary: During neuronal exocytosis, the vesicle-bound SNARE synaptobrevin 2 forms complexes with the plasma membrane-bound SNAREs syntaxin 1A and SNAP-25 to intiate the fusion reaction. SNARE proteins form complexes not only with each other, they also have been reported to homo-oligomerize, and they are known to have a multitude of binding partners. To study the spatial organisation of SNAREs and SNARE-interacting proteins in intact membranes we have recently established a procedure for the generation of surface-bound and inside-out sheets of plasma membranes from PC12 cells. These membrane sheets retain their capacity for regulated exocytosis and are accessible to external probes. We found that syntaxin is organised in cholesteroldependent microdomains that are distinct from detergent resistant membranes (rafts) and that define docking and fusion sites for secretory granules. The research program outlined here has two specific aims. First, we will exploit advanced confocal microscopy (4Pi microscopy) to determine the distance of docked granules to the syntaxin clusters with an accuracy of 10-20 nm and to correlate the distance with functional parameters. Second, we will investigate whether liposomes reconstituted with SNAREs and accessory proteins are capable of docking and fusion at such microdomains.
