Gab1 and Gab2 expressed in pancreatic beta cells were rapidly tyrosine phosphorylated in response to EGF stimulation. The result suggested involvement of Gab family adaptor proteins in EGF-driven beta cell proliferation. We explored a possible implication of Gab family in beta cell function and its proliferation by examining phenotype of double KO mice for Gab1 and Gab2. While beta-cell specific-Gab1/Gab2 double KO mice were successfully generated, their glucose tolerance and beta cell mass was comparable to those of control mice. These results suggested that no involvement of Gab family in regulation of beta cell mass. Other possibility may involve compensatory roles for other Gab family member, such as Gab3, in the regulation of beta cell mass.
