We have identified a novel gene termed ARIA that regulates angiogenic functions in endothelial cells. Here, we further characterized its function by using ARIA-deficient mice. ARIA-deficient mice demonstrated substantially enhanced neovessel formation comparing to WT mice assessed by hindlimb ischemia model. Moreover, we revealed that enhanced PI3K/Akt signaling in both endothelial cells and endothelial progenitor cells is attributable to the increased neovessel formation in ARIA-deficient mice. Also, we fond a small compound that potentially inhibit ARIA function by in silico screening. Inhibition of ARIA is a new therapeutic approach to treat ischemic diseases.
