Development of build-in adjuvant peptide vaccine bearing lipid A mimic

以脂质a （toll样受体4激动剂）的结构为基础，设计了一种用于内建佐剂亚单位肽疫苗的糖脂化合物，旨在通过与糖脂结合提高肽的抗原性。在固相肽合成过程中，将合成的糖脂与来自甲型流感病毒膜蛋白胞外结构域的M2e抗原通过连接物偶联，得到糖脂偶联的M2e作为内建候选疫苗。在不添加佐剂的情况下，通过小鼠免疫对糖脂偶联M2e进行免疫学评价。M2e特异性抗体是通过单独给药糖脂偶联的M2e诱导产生的，这意味着所设计的糖脂可用于内建佐剂疫苗的开发。

Aiming at improvement of peptide antigenicity by conjugation with glycolipid, a glycolipid compound was designed based on the structure of lipid A (Toll-like receptor 4 agonist) for applying to the build-in adjuvant subunit peptide vaccine. The synthesized glycolipid was conjugated with M2e antigen derived from the extracellular domain of influenza A virus membrane protein via a linker during solid phase peptide synthesis, yielded glycolipid-conjugated M2e as a build-in vaccine candidate. Immunological evaluation of the glycolipid-conjugated M2e was carried out by mice immunization without additional adjuvant. The M2e-specific antibody was induced by administration of the glycolipid-conjugated M2e alone, meaning the designed glycolipid was useful for build-in adjuvant vaccine development.
