Developing a new cell culture system for efficient production of infectious HPV virions to study molecular mechanisms of HPV infection cycles.

人乳头瘤病毒（HPV）是宫颈癌的一种病原，其生命周期依赖于宿主细胞分化，在实验室中阻碍了传染性HPV病毒粒子的产生。本研究的目的是开发一种新的细胞培养系统，以有效地生产感染性HPV病毒粒子，并研究HPV复制周期的分子机制。我们能够操纵细胞诱导HPV基因组扩增，并发现激活细胞DNA损伤反应，包括ATM和NF-κB可以抑制HPV基因组复制。

The life cycle of human papillomavirus (HPV), an etiological agent of cervical cancer, relied upon host cell differentiation, hindering production of infectious HPV virions in a laboratory. The aims of this research are to develop a new cell culture system for efficient production of infectious HPV virions and to study molecular mechanisms of HPV replication cycles. We were able to manipulate cells to induce HPV genome amplification and found that activation of cellular DNA damage response including ATM and NF-κB can suppress HPV genome replication.
