Entscheidende Schritte in der Lymphozytendifferenzierung werden durch die Wnt- und Notch-Signaltransduktionskaskaden gesteuert; dabei spielen Groucho-enthaltende Repressorkomplexe eine wichtige Rolle. Als Korepressor bindet Groucho nicht direkt an DNA, sondern wird von DNA-gebundenen Repressoren an Promotoren rekrutiert und kann daraufhin aktiv Transkription unterdrücken. Der starke repressive Effekt von Groucho wird durch mindestens zwei molekulare Mechanismen erzielt: Groucho kann sowohl mit Histon Deacetylasen als auch direkt mit der generellen Transkriptionsmaschinerie über den Mediator der transkriptionellen Regulation interagieren. Das Ziel des Projektes ist es, Groucho-enthaltende Komplexe aus Lymphozyten zu isolieren. Proteine, die zu diesem Schlüsselrepressorkomplex gehören, sollen daraufhin in vitro sowie in vivo in Hinblick auf transkriptionelle Regulation in der Lymphozytenentwicklung analysiert werden.
