本研究課題では、米油製造時に副生する脱脂米糠からタンパク質・有機リン化合物の双方を選択的に分離・精製可能なIP-EWTプロセスと脱脂米糠糖化物に含有するC5、C6糖から高不飽和脂肪酸含有油脂の生成が可能な微生物生成プロセスの双方を融合し、脱脂米糠からのタンパク質、有機リン化合物、油脂生産を可能にする脱脂米糠の完全再資源化プロセスの確立を目指した。これまでに、脱脂米糠糖化液を構成する糖質(グルコース、キシロース、アラビノース)に対する油性酵母の資化性、油脂生成能、生成する油脂の脂肪酸組成等の生物工学的基礎知見の蓄積が為されていない。そこで本年度(令和4年度)は、油性酵母の脱脂米糠糖化液を構成する糖質に対する菌体増殖および油脂生成能、生成油脂の脂肪酸組成を明らかにし、得られた各パラメーターを主成分分析(biplot)により、各菌株の油脂生産における特徴やが脂肪酸組成に与える各当成分の影響の理解を目指した。Rhodotorula sp.C7(C7株)、Rhodosporidium paludigenum C10(C10株)、Sporidiobolus pararoseus,、KX709872(KX株)の3株を用いた。油脂生産試験(好気-回分培養)の結果、C7株は、脱脂米糠糖化液を構成する糖質(単糖)を含む単一炭素源培地に おいて、3種それぞれの糖質に対する資化性が高く、3種の糖質を含有する脱脂米糠糖化液培地においても、優れた菌体増殖、油脂生産性示した。さらに、得られた培養結果を主成分分析(biplot)に供した結果、リノール酸及びリノレン酸含有率は菌体増殖、油脂・EPS生成量と負の相関関係を示すことが明らかとなった。
This research topic is about the separation and purification of the two kinds of organic compounds from defatted rice bran, which is a by-product in the production of rice oil. The two kinds of organic compounds from defatted rice bran can be separated and purified. IP-EWT can be used to prepare defatted rice bran saccharide containing C5 and C6 sugars, high unsaturated fatty acids, and oil. The production of microorganisms can be combined with the two kinds of organic compounds from defatted rice bran The aim of this paper is to establish a complete recycling system for defatted rice bran in the production of fats and oils. The accumulation of basic knowledge of biotechnology, such as the chemical properties of oily yeast, the ability to produce fat, and the fatty acid composition of fat produced by degreasing rice bran saccharification solution. This year (Y2K and Y2K), the study aimed to understand the effects of various components on the carbohydrate composition of the degreased rice bran saccharification solution of oily yeast, cell growth capacity, fatty acid composition of oil and fat production, and biplot analysis. Rhodotorula sp.C7(C7 strain), Rhodosporidium paludigenum C10(C10 strain), Sporidiobolus pararoseus, KX709872 (KX strain) and three other strains were used. The results of oil production test (good quality-subculture) showed that C7 strain had high productivity for sugar (single sugar) composition in defatted rice bran saccharification solution containing only one carbon source, three kinds of sugar, three kinds of sugar and defatted rice bran saccharification solution containing three kinds of sugar, excellent cell growth and oil production. The results of principal component analysis (biplot) showed that there was a negative correlation between cell growth, oil and EPS production and the content of organic acids.
