令和4年度は天然型基質を調製すると共に、市販の基質を用いた基質特異性の違いを明らかにする予定としていた。また、基質特異性の違いの鍵となる酵素の立体構造を明らかにすることを計画した。そのため以下の各項目について実験を進めた。① 針葉樹（スギ、カラマツ）由来の天然型マンナン系基質調製：イオン液体抽出法により修飾基との共有結合を保持した基質を調製する方法について試みており、その構造を解析中である。② ４種の菌由来のファイミリ―５に属する９種類の酵素の基質特異性の解明：遺伝子組み換えによる酵素の調整を行い、その一部について基質の特異性の解明を行った。③ 精製酵素の結晶化：得られた酵素の結晶化キットによる結晶の作成し、解析可能な闕所を得ることができた。④ X 線回折による立体構造の解析：高エネルギー研究所のビームラインを使って立体構造の解析を行った。以上の実験により、β-1，4結合の切断の選択性は高いものの、構成糖についてグルコースであるか、マンノースであるかの点においてはその認識があいまいな酵素が存在することを明らかにすることができた。引き続き構造面からその違いを明らかにするように継続して実験を行っている。
