日本では毎年5000人ものAYA世代の若者ががんに罹患している。AYA世代のがん対策は政策提言にもある通り、これからの日本の医療にとって大きな課題である。今回我々はAYA世代で問題となる希少がん、特に治療法の進歩の乏しい軟部肉腫に注目した。軟部肉腫の切除不能例に対する化学療法はDoxorubicin単剤が未だに主流であるが効果に乏しい。進行軟部肉腫の予後改善のため、新規治療法の開発がAYA世代のがん患者救済のためにも必須である。申請者らはこれまでに、Liposomeを利用した新たなDrug delivery system (DDS)を開発（Nat Biotechnol. 2008）。この技術を応用し新たなる標的抗がん療法を開発し特許も申請した（特許第5936002号）。そこで本研究ではこのDDSで制御性T細胞を標的とした新たな手法での免疫原性細胞死による新規抗がん治療法の開発をすることを目標とする。健常人ドナー血液からヒト末梢血単核球を比重遠心法にて分離した単核球を既報に従いsortingしたSortingした各分画のＴ細胞に標識し導入効率をflow cytometerで定量的に検討。BCL2阻害剤と結合したCD25標識-BCL2阻害剤含有Liposomeを作製。IL-2の濃度勾配をつけ（0, 1, 5, 10, 100, 1000 IU）刺激し、7-AADとanexin VによるApoptosis assayにて選択的な殺細胞効果を評価した。
