初年度の実験予定であるヒト大腸癌細胞株について、抗PK-R1(Prokineticin-receptor1)抗体ならびに抗PK-R2(Prokineticin-receptor2)抗体を用いた免疫組織化学染色法にて細胞における発現の局在、強度などを検討をおこない、その発現状況により高PK-R1発現型・高PK-R2発現型大腸癌細胞株の作製とPK-R1・PK-R2ノックアウト大腸癌細胞株の作製する細胞種の選定を終了した。また導入予定のPK-R1ならびにPK-R2発現ベクターは準備が整っており、前実験の検討において細胞内での発現も確認され、本実験に入っている。さらに高PK-R1またはPK-R2発現型大腸癌細胞株にsh(small hairpin)RNAを挿入して、ノックアウト大腸癌細胞の樹立の準備も進んでおり、次の段階に向かっている。
