研究目的のヒト廃棄胚を用いて、初期胚分裂過程の染色体分離過程に着目し、多重蛍光標識による動的挙動解析を行っている。良好な着床前胚発育のためには侵襲性が低い手法が必要であり、多重蛍光標識のための観察用培地内への胚浸染を行い、レーザー操作型共焦点顕微鏡を用いた画像取得を行っている。またヒト胚、マウス胚において得られた染色体分離過程の画像についてVisual Object Taggin Tool (VoTT)を用いた細胞内小器官部位に対するアノテーションを施行し、正倍数性、異数性発生に関与し得る異常分裂パターンに対する画像解析のための学習用データの作成を行っている。これらの過程においては種差、個体加齢の他、妊孕性温存療法においては、卵子凍結前後の処理・処置におけるタイムラグの影響も考慮する必要があり、われわれがこれまで検討してきた体外成熟培養(in vitro maturaion, IVM)および、第1極体放出から凍結までのタイムラグ、前核の出現から消失までのduration of pronucleus presence, DPPらをパラメータとし、これらの影響も含めた検討を行っている。IVM, 卵子凍結過程における上述のパラメータ検討の際にはタイムラプスモニタリングを用いた検討も必要であるが、各生殖補助医療施設のlaboratoryにおいて、施設の配置率、胚培養士などの人員配置は異なっているため、最適な卵子凍結の実施のためにはこれらの要因を含めた検討も必要であり、本邦の現状についても考慮している。
