まず、眼窩組織の冠状断での標本を作製し、平滑筋染色を行った。白人標本を用いた既報では、眼球赤道部レベルにおいて、内直筋から下直筋へかけて平滑筋線維が多数分布していたため、日本人においても同部位が染まると予測していたが、染まりが悪かった。そこで再染色を行ったが、同様に染まるが悪かった。そこで日本人においては白人と異なり、眼球赤道部レベルにおける内直筋から下直筋へかけての平滑筋線維の分布はほとんどないことが推測された。本研究における核心は、眼窩脂肪から平滑筋線維を単離し、筋線維芽細胞へ脱分化させることであったが、その根本である平滑筋線維の単離が困難であり、また仮に平滑筋線維の単離が可能であったとしても、それを筋維芽細胞へ脱分化させることも難しいことがわかった。そのため、本研究を中止することにした。
