近年,小腸上皮幹細胞から産生される多様な分化細胞を含み,消化管上皮のホメオスタシスをin vitroで再現する「オルガノイド」培養法の開発が加速している.当研究室では,絨毛構造を模したオルガノイド二次元培養法の構築に成功しており(Sugimoto et al. Nature. 2021),本研究は,この系をさらに改善し,消化吸収などの機能を有した,より消化管上皮らしいin vitro培養系を開発することを目的としている.さらに,この系を応用し,近年増加している炎症性腸疾患の病態を解明するため,消化管上皮の修復機構を検証するin vitro培養系への応用を目指す.今年度は,昨年度習得した絨毛構造を有するオルガノイド二次元培養法のさらなる改変を行うとともに,上皮細胞の機能解析へと段階をすすめた.培養条件については,昨年度成功に至った消化管上皮分化マーカーの発現誘導を,ヒト組織の発現レベルへと近づける条件を探索し,機能アッセイの結果と相互にフィードバックしながら改変を行っている.機能解析については,消化管の主要な機能である栄養素の吸収能に着目し,LC/MS解析によってグルコース(3-O-Methyl-D-glucose)の細胞内への取り組みを確認することに成功している.
In recent years, the epithelial cells of the small intestine have been divided into multiple cells, and the digestive tract epithelial cells have been detected again. The culture method has been used to speed up the development of in vitro. When the laboratory was in full swing, the animal model was established, and the two-dimensional culture method was successfully developed (Sugimoto et al. Nature. 2021), in this study, the clinical symptoms of inflammatory diseases have been improved, the digestive tract epithelium has been improved, the digestive tract epithelial in vitro has been improved, and the digestive tract epithelium has been opened. The purpose of this study is to improve the diagnosis of inflammatory diseases. The digestive tract epithelium repair machine is used in the in vitro culture system. This year, we learned that the two-dimensional culture method was used to improve the performance of the digestive tract epithelial repair machine, and the epithelial cell machine was able to analyze the segment of the digestive tract. The condition of the culture was very good, and it was successful in the last year that the differentiation of the digestive tract epithelium was induced. The organization is responsible for the exploration of the conditions for near-term health care, and the results show that there is a change in the operation of the machine. The machine is able to analyze the equipment, and the main organ of the digestive tract is capable of monitoring the absorption of nutrients. The LC/MS parsing method was used to determine the success of the intracellular DNA retrieval group (3-O-Methyl-D-glucose).
