【実験動物】マウスMHCのH-2領域のみの異なるC57BL/10(H-2b),C57BL/10.D2(H-2d),C57BL/10.BR(H-2k)の3種類のB.10コンジェニックマウスを用いた.【実験方法】マウスの腹腔内にStaphylococcal enterotoxin B(SEB)をミニ浸透圧ポンプにより10g/day投与し,3週間投与後に以下について検討した.1)脾細胞および2)胸腺細胞リンパ球サブセット,3)腎組織所見.Controlとして,生理食塩水を投与したマウスに対して同様の実験を施行した.【結果および考案】1)脾細胞リンパ球サブセット:C57BL/10においては,全リンパ球中のTリンパ球(TCRβ陽性細胞)の割合がSEB投与群において有意に増加していた.また,Vβ3陽性Tリンパ球,およびVβ7陽性Tリンパ球においてはSEB投与群とControl群において有意差は認められなかったものの,Vβ8陽性Tリンパ球の割合はSEB投与群において有意に増加していた.C57BL/10.D2およびC57BL/10.BRにおいては,全リンパ球中のTリンパ球,Vβ3陽性Tリンパ球,Vβ7陽性Tリンパ球,Vβ8陽性Tリンパ球の割合ともSEB投与群とControl群において有意差は認められなかった.2)胸腺細胞リンパ球サブセット:C57BL/10,C57BL/10.D2およびC57BL/10.BRの3群において,全リンパ球中のTリンパ球,Vβ3陽性Tリンパ球,Vβ7陽性Tリンパ球,Vβ8陽性Tリンパ球の割合ともSEB投与群とControl群において有意差は認められなかった.3)腎組織学的検討:C57BL/10,C57BL/10.D2およびC57BL/10.BRの3群において,蛍光抗体法による検討上,メサンギウム領域優位のIgG,IgMの沈着が,Control群と比較してSEB投与群でやや強い傾向が認められたものの,IgA,C3の沈着の程度は2群間に有意差は認められなかった.また,光学顕微鏡による検討では有意な病変は認められなかった.今後は,SEBの投与期間を延長して同様の実験を試み,血清および腎組織上でのサイトカインの検討も加える予定である.

C57BL/10 (H-2b), C57BL/10.D2 (H-2d), C57BL/10.BR (H-2k) and B.10 (H-2d) are three species of MHC. [Methods] The intraperitoneal administration of Staphylococcus enterotoxin B (SEB) at 10 g/day for 3 weeks was followed by the following examinations: 1) Splenocytes; 2) Thymocytes; 3) Kidney tissue findings. 1) Spleen cell differentiation: C57BL/10, TCR β positive cells, TCR β positive cells, and SEB cells were intentionally increased. V β 3-positive T release ball, V β 7-positive T release ball, V β 8-positive T release ball, V β 7-positive T release ball, V β 8-positive T release ball, V β 8-positive T release ball, V β 7-positive T release ball, V β 8-positive T release ball, V β 7-positive T release ball, V β 8-positive T release V β 8 positive T cells, T cells, T cells C57BL/10, C57BL/10.D2 C57BL/10.BR V β 8-positive T C57BL/10, C57BL/10.D2, C57BL/10.BR and C57BL/10.BR are three groups in which IgG, IgM and IgM are present in the region of C57BL/10, C57BL/10.D2 and C57BL/10.BR. Control group comparison between SEB and SEB groups is a strong tendency to recognize IgA, C3 and the degree of calm is a significant difference between 2 groups. The optical microscope is designed to detect the presence of a virus. In the future, SEB will be administered for a longer period of time.
