本研究は、成長期男女のスポーツ選手における運動・トレーニング量(長期)及び一過性の持久的運動(短期)に対するストレス耐性を、DNA 損傷・修復能、DNAテロメア長、心肺持久力(体力レベル)及びエストロゲン分泌量の相互関連性から検討することを目的とした。研究方法として、第二次急伸期後の女子中学・高校生(15~18歳)及び男子中学・高校生(15~18歳)を対象に、自転車エルゴメーターでの最大運動負荷試験を実施し、心肺持久系能力を測定した(換気性作業閾値、呼吸性代償及び最高酸素摂取量: VO2peakの測定)。また、最大下運動(VO2peakの65%の運動強度: 65%VO2peakで60分間の自転車運動)を実施し、運動前後で唾液(運動前後10分間)及び尿検体(2時間尿:運動前-2~0時間、運動直後0~2時間、運動後24~26時間)を採取した。特に、女子中学・高校生においては、エストロゲン(17β-estradiol)濃度が異なる卵胞期と黄体期で、最大下運動をそれぞれ1回ずつ(計2回)実施した。令和4年度(2022度)の生化学分析として、中強度持久的運動後のDNA損傷及び修復能に関する性差の影響を調べ(尿中8-hydroxy-2’-deoxyguanosine: 8-OHdGの定量)、体力レベルを同等に比較した場合の成長期男女における中強度持久的運動後のDNA損傷及び修復能は類似しているという結果を得た。その結果に準じて、現在、尿中ステロイドホルモンレベル(エストロゲン)、唾液に基づくDNAテロメア長及びミトコンドリアDNAコピー数を定量中で、段階的に各種生体マーカーにおける相互の関連性を特定する予定である。
The purpose of this study was to investigate the correlation between physical activity (long-term) and transient long-term exercise (short-term) in male and female athletes during growth, and the relationship between physical tolerance, DNA damage, DNA repair, cardiopulmonary endurance, and secretion. Methods: The maximum exercise load test was conducted among girls 'high school students (15 - 18 years old) and boys' high school students (15 - 18 years old) after the second stretch period, and the cardiopulmonary endurance capacity was measured (the threshold value of metabolic work, respiratory compensation and maximum acid content: VO2peak). The maximum downward movement (VO2 peak and 65% of exercise intensity: 65% VO2 peak and 60 minutes of bicycle movement) was performed, saliva was taken before and after exercise (10 minutes before and after exercise) and urine was taken for 2 hours (urine: -2 to 0 hours before exercise, 0 to 2 hours after exercise, 24 to 26 hours after exercise). In particular, girls 'high school and college students, the concentration of 17β-estradiol varies from egg stage to luteal stage, and the maximum downward movement is 1 cycle (counting 2 cycles). The biochemical analysis of DNA damage and repair after moderate and prolonged exercise in the fourth year (2022 ℃) was conducted to adjust the effect of sex difference (quantitative analysis of 8-hydroxy-2'-deoxyguanosine: 8-OHdG in urine) and physical strength. The results showed that DNA damage and repair after moderate and prolonged exercise were similar in men and women at the same growth stage. The results were accurate, present, urine, saliva, DNA, DNA length, and the number of DNA samples quantified, graded, and interrelationships among various organisms determined.
