项目摘要

本研究は抗HIV薬投与下の細胞傷害性Tリンパ球（CTL）反応誘導がウイルスへ及ぼす影響について広く解析することを目的としている。本年度は研究計画段階において予定されていたCTL反応のウイルス複製抑制能評価に加え、昨年度実施したウイルスゲノム解析の結果を元に、ウイルス複製へ影響を及ぼした抗原特異的CTL反応のエピトープ探索を行った。投薬前、投薬下、投薬中止後のCTL反応のウイルス複製抑制能評価から、投薬下のCTL反応誘導によってウイルス複製抑制能は増強されうることを明らかにした。また、昨年度に解析した抗原特異的CTL反応との関係について解析し、特にGagを標的とするCTL反応頻度とCTL反応誘導後のウイルス複製抑制能との間に相関関係があることを示した。さらに、同じく昨年度に解析した血漿中ウイルス量との関係についても解析し、投薬中止後のウイルス量がCTL反応誘導後のウイルス複製抑制能と逆相関の関係にあることを明らかにした。これらのことは、Gag特異的CTL反応のウイルス複製抑制への寄与を示唆する重要な結果である。また、昨年度に行った投薬前後のウイルスゲノム解析結果から、特にウイルスの複製に影響を及ぼしたと考えられるCTL反応の抗原エピトープ探索を行い、ワクチン抗原であるGagおよびVifの一部を標的抗原エピトープとするCTL反応が、主要組織適合遺伝子複合体クラスIのハプロタイプを共有する一部個体において、共通して誘導されていることを確認した。本研究で得られた結果は、HIV感染症においてウイルス複製抑制に有効なCTL誘導法の開発に結び付くことが期待される。
