A DIFFRACTOMETER SYSTEM FOR PROTEIN CRYSTALLOGRAPHY

Funds are requested for the purchase of an x-ray diffractometer system that will provide the primary crystallographic data for a series of interdisciplinary investigations of enzyme structure and function. Crystal structure analyses of flavodoxins, of complex flavoproteins in electron transfer chains, of flavo protein oxygenass, of Fe- and Mn- dismutases, and of Fe+3 dependent dioxygenases, conducted as an integral part of the ongoing projects of six faculty and two primary research scientists, will depend on the instrument for accurate measurements of diffracted intensities. Current and projected investigations encompass analysis of the enzymes themselves, of their complexes with substrates, products, and inhibitors, and of forms that accumulate when the reaction cycle is stopped at low temperatures. In addition, enzyme structures modified by the introduction of altered co-factors and by site-directed mutagenesis will be examined. The diffractometer system will include an x-ray generator, a four-cycle goniostat, a driver computer with accessory disks and associated software, a tape drive, a terminal, and low temperature accessories.
