DNA-encoded libraries provide an exciting new technology for finding start points for drug discovery. They consist of a collection of small molecules, covalently attached to a unique DNA-tag that is specific to each compound, enabling efficient protein target screening. Despite the current promise offered by DNA-encoded libraries, many traditional methods of organic synthesis are not compatible with DNA-tagged substrates, for example, acidic and oxidising conditions which cause DNA damage. This project will explore the scope for recently developed micellar methodology to facilitate these traditionally forbidden reactions, by localising the organic reactants to the substrate portion of the molecule and protecting the DNA-tag from the reaction conditions.
