This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The goal of this project is to become familiar with the technical aspects of labeling zebrafish embryos that express proteins genetically altered to contain a tetracysteine tag. This tag is recognized by the commercially available fluorophores FlAsH-EDT2 and ReAsH-EDT2. Binding of either fluorophore to the tetracysteine tag results in a fluorescent signal that can photoconvert diaminobenzidine into an osmiophilic precipitate.
