TWO-HYBRID SCREENING WITH POTENTIAL CHROMOSOME SEGREGATION PROTEINS

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. Our lab studies chromosome segregation in budding yeast with a focus on the kinetochore which is a large protein complex that mediates interaction between DNA and the spindle. I have previously collaborated with the Yeast Resource Center to perform two-hybrid screens with kinetochore proteins, which resulted in a publication. I have continued my studies of the kinetochore in my own lab, with a focus on the Ndc80 complex, which is a highly conserved complex that is required for spindle checkpoint function. My lab has taken a biochemical and genetic approach to identify genes and pathways that interact with the Ndc80 complex. We have identified two genes that may have a role in kinetochore function and would like to identify interacting proteins using genome-wide two-hybrid screening.
