The spindle checkpoint regulates the metaphase-anaphase transition, and aims to prevent aneuploidy.It responds to defective kinetochore-microtubule interactions by delaying sister chromatid separation and anaphase onset. In so doing it prevents chromosome mis-segregation and the generation of aneuploidy. Aneuploidy is lethal in single-celled organisms such as fission yeast, and is one of the hallmarks of cancer in humans. We are using fission yeast as a model system to study the in vivo dynamics and interactions of the Mad and Bub checkpoint proteins.
