This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. PH domain leucine-rich repeat protein phosphatase (PHLPP) terminates growth factor- and G-protein coupled receptor (GPCR)-induced signaling through dephosphorylation of the hydrophobic motif site in the carbolxyl-terminus the AGC kinases, Akt and PKC. PHLPP knockdown increases Akt activity towards a variety of substrates including TSC2, FOXO and GSK3 and reduces apoptosis in response to etoposide. We are currently testing the role of PHLPP in autophagy.
