This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Our goal is to investigate the effect of advanced age on antiepileptic drug (AED) pharmacokinetics and metabolism. Selected elderly and adult male and female ambulatory patients taking phenytoin (PHT) or carbamazepine (CBZ) for any reason as monotherapy or with other non-interfering medications including other non-interfering AEDs will be evaluated. The patients will receive a portion of their oral dose of CBZ or PHT in a stable-labeled formulation given intravenously as 100 mg over 10 minutes (one time only). Stable-labeled CBZ and PHT are NOT radioactive and exhibit the same kinetics and metabolism as unlabeled CBZ or PHT. Blood samples and urine collections will be analyzed. In the CBZ De-Induction Study, a sub-study of the CBZ Project, subjects receive the intravenous formulation at four time points over three months while being taken off their CBZ regimen and switched to another antiepileptic drug (AED).
