This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The goal of this pilot study is to determine the lipid phenotype, SR-BI (scavenger receptor class B type I, HDL receptor) genotypes, and evidence for subclinical atherosclerosis in infertile women. These investigators have recently shown that infertile women with SR-BI RNA levels below the 25th percentile had significantly lower baseline and peak estradiol levels and lower number of retrieved and fertilized oocytes compared to women with SR-BI RNA levels above the 75th percentile. Now they seek to determine the prevalence of SR-BI variants in this population and the association of these variants with the lipid profile, subclinical atherosclerosis and fertility measurements. There are 3 specific aims: 1) To measure the lipid phenotype in infertile women and assess the ability of monocyte-derived macrophage and granulosa cells isolated from these subjects to mediate selective cholesteryl ester uptake from HDL (a functional test of SR-BI, which presumably would be impaired in low expressers compared to high expressers).; 2) To associate alterations in SR-BI expression, function and gene variants with pregnancy outcome measurements and atherosclerotic risk (assessed by intimal wall thickness on carotid doppler) in infertile women. 3) To identify variants within the SR-BI gene in infertile women.
