This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
This two-year prospective study uses a noninvasive imaging technique called blood oxygenation level-dependent magnetic resonance imaging (BOLD MRl). We plan to determine (1) whether chronic kidney disease (CKD) and chronic allograft nephropathy (CAN) are associated with poor kidney oxygenation (2) whether poor oxygenation results in increased oxidant stress (OS) in CKD and CAN (3) whether losartan, an angiotensin receptor blocker, has anti-oxidant properties in the kidney. The study will include a group of healthy subjects and two groups of patients (CKD and CAN). Subjects will be scheduled for two visits (24 months apart), where blood and urine samples will be collected before and after a 50 mg oral dose of losartan. Individuals will also undergo BOLD MRI testing of their kidney before and after losartan. Urine and blood samples will be examined to measure kidney function and various markers of OS.
