This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
Wzz protein is a chain length determinant protein involving in O-antigen component of lipopolysaccharide (LPS) biosynthesis, and also behaves as a polymerase by incorporated various length of repeated O-units (O-antigen), whose concept is correlated to our proposed studies for DNA polymerase. Various other biophysical methods (crystallography, NMR and Cryo-EM) have been attempted in the past years to obtain the structural information of this membrane protein wzz (Guo, H. et al. (2005) Appl. Environ. Microbiol., 71, 7995-8001). We have collaborated since three months ago with Prof. George Wang at the OSU regarding obtaining its structural information by SAXS. While no significant progress has been made by other methods, we have some very positive preliminary studies for this protein by SAXS. We would like use this rapid access for obtaining more preliminary SAXS studies before we submit our full proposal. Two papers regarding the preparation of this protein and biochemical studies have been published. Different length of wzz proteins, which are proposed to be correlated to different biological functions, will be available for SAXS studies.
