This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Activation of the protooncogene homolog Ras in yeast results in activation of protein kinase A (PKA). No assay currently exists to directly monitor activation of PKA in vivo. We propose to test whether an in vivo FRET assay based on a method developed by Roger Tsien and his colleagues for use in mammalian cells will function in yeast. The availability of a FRET assay for PKA activation would greatly facilitate physiological studies on nutrient signaling in yeast.
