We performed RNA sequencing (RNA-seq) of the EVs in the serum of HC (Healthy donors), MCI (mild cognitive impairment) and AD (mild dementia due to Alzheimers disease) individuals with 110 bp paired-end directional reads. We observed significant differences in mitochondrial RNA abundance in EVs from AD/MCI/HC serum. We validated these changes using RT-qPCR analysis and obtained plasma samples from an additional 5 individuals per group and performed RT-qPCR analysis. In summary, in EVs of MCI and AD patients, we found the levels of mt-RNAs are robustly and significantly more abundant. We are currently working on the metagenomic sequencing and corelating our data with the results of DTI.
