The Glucagon-subfamily receptors (GSRs) belong to the largest mammalian receptor family of G-protein coupled receptors (GPCRs). Most notably, the glucagon-like peptide 1 receptor (GLP1R), the Glucagon-like peptide-2 (GLP2R) receptor and the glucagon receptor (GcgR) are important for maintaining blood sugar levels in animals. The hormone GLP1, which binds to and activates the GLP1R, is responsible for glucose stimulated insulin secretion from pancreatic insulin producing cells to reduce blood sugar. In contrast, the major action of glucagon is to act through its receptor (GcgR) in the liver to increase glucose production to alleviate or prevent blood sugar from falling too low. GLP2 is found in mainly in intestinal cells and regulates among other things sugar absorption into the bloodstream. The activation of these GSRs like many other GPCRs, is mediated through accessory/interacting proteins, however for most receptors, these remain unknown and undiscovered.
