生物体的一个基本功能是感受摄入的营养类型和数量,进而调节自身的代谢状态。因此发现特定营养的感受原理及其生物效应的分子机制,不仅可以理解营养感受细胞和效应细胞的工作原理,也为特异地改变代谢状态,进而调控生理和病理过程提供了可能。本课题组通过前期实验,发现果蝇脂动激素(Akh)分泌细胞是一个之前未被认识的氨基酸感受器。这些细胞被特定氨基酸激活后释放Akh(胰高血糖素类似物),进而诱发脂肪体细胞的钙离子震荡和代谢变化。我们还发现,苏氨酸与甲硫氨酸对于快速发育中的果蝇幼虫具有依赖于Akh信号的生物毒性,这一发现显示氨基酸感受通路可能参与了特定营养对发育的调控。本项目拟在现有发现基础上,进一步阐明Akh分泌细胞感知特定氨基酸的分子机制,明确Akh信号对脂肪细胞氨基酸代谢的影响,并揭示特定氨基酸产生激素依赖的发育毒性的原因。本研究将从多细胞通信的角度促进对生物体氨基酸感知及其调控作用的深入理解。

One fundamental function of most living organisms is to sense the type and quantity of nutrients ingested, such that to further regulate their metabolic state. Therefore, understanding the molecular mechanism of how a specific nutrition can be detected and its subsequent metabolic effects, will not only provide knowledge about the mechanisms of how nutritional sensor and effector cells function but also offer opportunities to precisely manipulate the metabolic states and to control different physiological and pathological processes. Through functional live-cell imaging of fruit fly larvae, our team found that Akh-producing cells are a previously unidentified amino acid sensor. These neuroendocrine cells can be activated by specific amino acids, then release a peptide hormone Akh, a functional analogue of human glucagon, which will induce Ca2+ oscillation and metabolic change in the fat body cells. We further found that particular amino acids (threonine and methionine) show significant Akh-dependent toxicity in the fast-growing fly larvae, suggesting that this amino acid-sensing mechanism may be involved in the nutrient-dependent development regulation. Based on our current findings, we will investigate: the molecular mechanism of how Akh-producing cells perceive specific amino acids, the effect of Akh signaling on the amino acid metabolism in adipocytes, and how specific amino acids generate hormone-dependent developmental toxicity. This study will provide in-depth knowledge about the perception and biological effects of amino acid from an intercellular communication perspective.
多细胞生物的基本功能之一是感知体内的营养变化,进而调控细胞的代谢状态,以保持自身稳态。而代谢状态的改变不仅可以影响发育和衰老过程、调控生理机能、诱导睡眠、进食、繁殖等行为,也和肥胖、糖尿病以及肿瘤等病理状态密切相关。因此从根本上理解生物体对物质和能量的感知和调节机制,可以为防治人类疾病提供重要的理论基础。目前对特定细胞内的代谢调控网络已经有了较为深入的认识,然而高等动物的代谢调节往往并非发生在单一细胞类型中,而是利用不同细胞作为营养感受器和代谢效应器。因此,当前代谢领域的一个难点和热点是如何研究不同类型的细胞间的相互通信协作。黑腹果蝇作为一个优秀的研究多种细胞相互通信的遗传模型,具有与人类高度相似的代谢调控信号以及更加简单、经济、快捷的特点,因而日益受到代谢研究者的重视。本课题组通过前期的活细胞成像实验,发现果蝇脂动激素(Akh,人类胰高血糖素的类似物)分泌胞是一个之前未被认识到的氨基酸感受器:它可以接受特定氨基酸刺激并激释放脂动激素,进而调节脂肪细胞的代谢水平。本课题通过深入研究这一新的神经-内分泌-脂肪代谢调节通路,发现神经元感受特定氨基酸的分子机制,脂动激素调节脂肪细胞氨基酸代谢的分子机制,以及器官水平的钙波动激发的分子机制。本课题在方法学上结合了遗传学和活细胞成像技术,通过实时记录生物体内不同细胞的状态来研究营养感知和调控。相比传统的生化手段,我们可以直接获得代谢过程中精确的时间和空间变化规律。从研究模型角度,本课题提出了一个新的果蝇氨基酸感知-代谢调节通路,可以为深入理解氨基酸调控的胰高血糖素分泌机制及其生物学效应提供有效的遗传实验模型。
